THE DEVELOPMENT OF GLUCOSAMINE ANALYSIS IN THE SHARK CARTILAGE USING HIGH PERFORMANCE LIQUID CHROMATOGRAPHY

Abstract
The study is to analyze the development of glucosamine in the shark cartilage by employing High Performance Liquid Chromatography (HPLC) with UV-Vis detector. The analytical method is done by using the derivatization process considering the fact that glucosamine is a polar compound and has no sufficient chromophor to absorb UV radiation. The research is conducted through steps of shark cartilage mineral processing, that is, the preparation of standard and sample solution, the determination of optimum conditions, the validation of analytical methods, and the analysis of glucosamine in the shark cartilage respectively.
The validation of the analytical method is shown by these results: correlation coefficient of linearity test 0.995, variation coefficient of precision test 0.38% - 1.42%, recovery of accuration test 98.24 % - 101.95 %, resolution test (Rs) 1.975, LOD 0.24 ppm, and LOQ 0,80 ppm. The concentration of glucosamine in the shark cartilage is 1.08% at 240 nm UV detector, a C18 column (Lichocart (R), 250 mm x 4 mm), mobile phase aqua bidestilata: glassial acetic acid: methanol (89.96:0.04:10 pH 2.67) and a flow rate 1.2 ml / min.
