Formulation and development of Sumatriptan loaded S-SMEDDS using piperine for synergistic effect on permeation

Abstract
The present investigation was undertaken with an objective to enhance the permeation of sumatriptan succinate from self microemulsifying drug delivery system (SMEDDS) using piperine as the permeation enhancer. On screening of SMEDDS componentsclove oil (Oil), Capmul MCM (Surfactant) and PEG 400 (co surfactant) showed highest solubility of Sumatriptan succinate.Optimization was carried out by mixture design and from that the best batch was SS7 having 28% oil, 25.5% surfactant and 26.5% co surfactant. This batch showed lowest globule size 156 and highest %T 99.1%.The optimized formulation was then solidified by using adsorbent neusiline US2 showed free flowing powder. The micromeritical properties showed good flowability and reconstitution study of the powder showed the prepared emulsion was showed milky emulsion appearance. In-vitro dissolution and permeation study of optimized formulation showed that piperine increase the bioavailability of drug as compare to S-SMEDDS loaded with drug and marketed formulation.
