Assessment of the Partial Purified Lipid a Extracted from Proteus mirabiliscell Wall as Adjuvant Immunotherapy

Abstract

P. mirabilis is considered one of the nosocomial infection causative agent, and its importance is related with the wide separation in different ecosystem, in addition to its relationship with renal and bile calculi, and because of an increase of bacterial resistance, this study goal to find substitutional substance used to treat bacterial infection through using natural biomaterial, thus using LPS (Lipid A moiety) as adjuvant immunotherapy by different routes of administration (Oral, intraperitoneal, and subcutaneous) and find the best one. The results showed, recovered lipid A was obtained from both collected LPS of pathogenic P. mirabilis isolate and standard bacteria approximately represented 20 % of LPS amount, with LD50 0.6 g/ml for the pathogenic one. The estimation of some immune parameters such as IL-1 showed i. p administration was led to a significant elevation of this interleukin in compared with other routes and control groups. Meanwhile, IL-2, IL-10, and IFN- percentages showed a significant elevation in all treated groups in compared with control. While TNF- percentage revealed variation in levels according to the route of administration, simultaneously both intraperitoneal and subcutaneous routes showed a significant elevation. Whereas the IgG level exhibited, no significant alteration observed with any treated groups in compared with control one.
