REGENERATION OF MULTIPLE SHOOT FROM CALLUS IN BOUCEROSIA TRUNCATOCORONATA (SEDGW) GRAVELY & MAYUR

Abstract

Plant regeneration of an important endangered medicinal plant B.truncato-coranata was achieved through callus regeneration. Culture medium consist of Murashige and Skoog (MS) medium with 2.22 to 13.32 μM BAP, 3.35 μM NAA and 3% (w/v) sucrose with pH adjusted to 5.8. After 8 days of culture at 24±2º C, the stem produced friable dark green callus in 11.10 μM BAP and 5.35 μM NAA. Calli of 0.3 -0.6 mm size were transferred t MS medium with same concentration of BAP and NAA for regeneration. The calli developed shoot buds after 4-5 weeks and the frequency of calli forming shoots varied from 20 % to 90%. Optimum shoot regeneration occurred on MS medium supplemented with 11.10μM and 5.35 μM NAA. On this medium 90% culture responded with an average number of 12-15 shoots per callus. Whole plants recovered following rooting of shoots in MS medium supplemented with 5.35 μM NAA. The plantlets were hardened off and more than 96% of survived plants were released to the shade house.
