Abstract
Today even after fifty years of its manufacture the decision to use clozapine still represents a challenge to the clinician because of the complexities involved in accurately assessing its risks and benefits. Debate and uncertainty as to what represents an adequate trial, indications, and the mechanism of action of clozapine is continuing. The author conducted a review of studies that highlighted its historical aspect, the mechanism of action, efficacy, tolerability, and indications of clozapine in current perspective.
