PHARMACOLOGICAL BASIS OF APPLICATION OF “CANEPHRON N” IN THE TREATMENT OF PATIENTS WITH CHRONIC PROSTATITIS AND BENIGN PROSTATIC HYPERPLASIA

Abstract

We have significant experience of effective application of «Canephron N» in men with chronic prostatitis (CP) and benign prostatic hyperplasia (BPH). However, the mechanism of this phenomenon is not studied. Nevertheless, the critical analysis of literature and clinical data gives an opportunity to make the conclusion, that intimate molecular mechanisms of clinical effectiveness of doxazosin and tadalafil, which are used traditionally in men with CP and BPH, still remain incompletely understood. In 2016 – 2017, we were the first to estimate that the alpha-1- adrenoblocker doxazosin and the inhibitor of phosphodiesterase 5 type tadalafil have one more else mechanism of action: they raise the rate of oxygen consumption by the tissue of prostate. We used the method of polarographic analysis in the regime of chronoamperometry to define the rate of oxygen consumption by homogenates of prostate tissue in rats. We supposed that the mechanism of Canephron's effectiveness in men with BPH and CP is the same. The aim: to study how does Canephron impact on the rate of oxygen consumption (ROC) by the homogenates of rats' prostate. Materials and methods: 0,5 liter of distilled water (temperature = 36,6 °С) was poured into two plastic cans. Every can was connected to polarographic device by the pair of electrodes. 30 male rats were used. Under anesthesia the prostate was resected from rats and divided into two parts with equal weight. Homogenate was prepared from every part. Further, every homogenate was placed into the cans. The air-tightness was created. 2 Canephron dragee were submerged into the first can and none into the second can. The circuit was closed under the voltage of 0,95 V. Current intensity (CI) was the same in both cans in the beginning of every experiment – 1.8 microampere. It was made the synchronous comparison between the CI in two different cans with water in each moment of time. The Wilcoxon's criteria was used. It was detected that CI decreases much faster and more remarkably in those cans, where before homogenate immersion Canephron was placed. Conclusion: Canephron raises the rate of oxygen consumption by the tissue of prostate. In such way, Canephron helps the prostatic tissue to consume О2 supplied by blood. We consider that this is the pharmacologial reason and the basis of application of Canephron in men with chronic prostatitis and BPH
