DIFFERENTIATION AND IDENTIFICATION OF DIFFERENT GENOTYPES OF SUGAR BEET (BETA VULGARIS L.) USING DNA MARKERS

Abstract

It is studied the genetic polymorphism of nine different genotypes of sugar beet (Beta vulgaris L. ) of Ukrainian and foreign selection using RAPD- and SSR- analysis. In the total sample group of the studied genotypes it was found and analyzed 14 alleles with four RAPD-markers and proved heterogeneity of materials according to allele frequency (0.22-0.67), indicating the possibility of their use for genotyping varieties and hybrids of sugar beet and determining the degree of genetic kinship between them. When using DNA-marker GZM 086 identified five alleles, due to which they were identified polymorphic hybrids of Ukrainian breeding of Verhniachka CHS 37, Віla Tserkva CHS 57, Ukraina CHS72, 64 one-seeded Yaltushkivskyi variety, and hybrid Katiusha of foreign selection. The identified, alleles can be used to build DNA profiles that characterize the genetic diversity of different genotypes of sugar beet. They were determined the genetic distances between genotypes studied in the sample according to their similarity or distance. It was found that the most genetically similar are hybrids (variety), belonging to the same cluster according to used DNA-markers, and remote according to the studied locus - with values of genetic distances, which approaches one. For identification and differentiation of varieties and hybrids of sugar beet it is appropriate to use the definition of intervarietal polymorphism using different DNA-markers
