Abstract

Among the various routes of drug delivery, the oral route is perhaps the most preferred by patients and clinicians alike. Ketorolac is currently administered intramuscularly (30 mg) and orally as conventional tablet (10 mg) for short-term management of post-operative pain and moderate to severe pain. It is a non-steroidal antiinflammatory cyclooxygenase inhibitor. It acts by inhibiting the synthesis of prostaglandins. The major side effects of Ketorolac are gastric mucosal erosions, ulcers and gastric bleeding. The aim of this study was to prepare a new mucoadhesive tablet formulation of Ketorolac Tromethamine in view of attaining prolonged effect of drug for better therapy with reduced dosing frequency. In present work, an attempt has been made to formulate buccoadhesive tabletof model drug and preparation of tablets using hydrophilic polymers like HPMC K15M, HPMC K4M and carbopol934. The buccal tablets were characterized on the basis of their physical parameters (hardness, thickness, weight variation) drug content, surface pH, swelling index, mucoadhesive strength, in vitro drug release were studied.
