OBTAINING AND APPLICATION METHODS OF GARANIUM PA-LUSTRE PHYTOCOMPLEX FOR STOMATITIS TREATMENT

Abstract

During recent years despite the significant advances in creation of chemical pharmaceuticals, the popularity of phytotherapy has grown greatly. Phytomedicines contain a large number of biologically active substances, which, compared with synthetic drugs, facilitate a wide range of pharmacological effects. Moreover, the advantages of phytoremedies are mild therapeutic effect that develops gradually, almost absence of contraindications and side effects, low toxicity, inability to induce addiction and the possibility of prolonged use in different age groups. Our attention was paid to Geranium palustre L., which is widespread, has been used as home-made medicine and is a cheap source of plant raw material. Today, it is known from literature sources that various types of the genus Geranium have astringent, anti-inflammatory, hemostatic, antimicrobial, antioxidant effects and reveal immunomodulatory properties. The proposed method of obtaining the phytoextracts from Geranium palustre herb consists in conducting multiple extraction of plant material for providing full extraction of active substances and high yield of the prepared product with a pronounced anti-inflammatory activity and conservation of the obtained plant complex of biologically active substances in the form of dry extract. For confirmation of anti-inflammatory activity comparative studies of phytoextracts, obtained by the proposed method, were carried out on the model of carragenin edema of the feet in rats in comparison with known anti-inflammatory drugs ketanol and diclofenac. For a clearer picture of the action of the proposed extract of Geranium palustre it was decided to check its effectiveness in the treatment of inflammatory diseases of the oral mucosa, in particular, in the treatment of stomatitis. For this purpose, the obtained by described method phytoextracts of Geranium palustre were applied to a dental film that firmly adheres to the surface of the mucosa of oral cavity and quickly resorbates (up to 10 minutes), providing release of the plant remedy directly into the lesion area, and, thereby, achieving the maximum therapeutic effect. Experimental model of ulcerative stomatitis was obtained in the rabbits by a burn. Animals of the experimental group after the development of experimental stomatitis were treated by applying to the wound a stomatologic film with extracts of Geranium palustre herb. Visual monitoring of the condition of simulated ulcerative stomatitis in the region of the lower lip was performed in accordance with the experimental schedule (3rd, 5th, 7th and 10th day). The criteria for evaluating the effectiveness of local treatment were the timing of the elimination of perifocal inflammation, hyperemia, infiltration of the edges of the inflammation center, cleaning of the wound from necrotic tissues, the beginning of marginal epithelization and the time of its completion. It was found that during treatment using phytoextracts wound healing was faster, regenerative processes were more intensive, and signs of inflammation, that had occurred in the area of the damaged gum mucous membrane, decreased in experimental animals. As a conclusion, it is proposed an im-proved method for obtaining a standardized stable lyophilized phytoextract of Geranium palustre herb with high content of biologically active substances, which has a pronounced anti-inflammatory activity and can be used for creation on its basis the effective drugs with anti-inflammatory action for the symptomatic and etiological treatment of the wide range of diseases, including stomatitis.
