Abstract
Success of PCRbased molecular method is highly dependent on quality extraction of DNAtemplate. There are many methods currently available for DNApurification fromsemen;but,problemsrelatedtocontamination with foreign DNA, PCRinhibitors,andsusceptibilitytofragmentation are very common. In the present study, five different DNAextraction procedures were examined to ascertain their relative effectiveness for extracting DNAfrom semen samples of buffalo bull. The five methodsincluded-twocommerciallysuppliedkits(QiagenDNeasyBloodandTissueKit,PurelinkInvitrogen),modifiedQiagenDNeasyBloodandTissue Kit method, Chelex 100 method andPhenol-chloroformwithmodifiedlysisbuffermethod.Thequalityandquantity assessment of the differentially extractedDNArevealedsignificantdifferences among the five procedures. Modified Qiagen DNeasy Blood and Tissue Kitmethodwasfoundtobemostappropriate for extracting high quality and suitable quantity of DNAfrom semen of buffalo bull.
