Abstract
The aim of this study was to prepare Terbinafine hydrochloride nail lacquers for the treatment of Onychomycosis using permeation enhancers and demonstrate the effect of a novel permeation enhancer system by in vitro studies. Nail lacquer formulations were prepared by simple mixing method with varying concentration (1% to 3% v/v) of permeation enhancers (Thioglycolic acid and DMSO) as alone and in combination. Terbinafine hydrochloride was selected as a model drug which is an allylamine antifungal drug. The lacquers were compared for glossiness, drying time, smoothness of flow, non volatile content and water resistance. The in vitro studies were carried out on artificial membrane (phosphate buffer pH 7.4 and methanol AR grade, in the ratio 4:1) using a Franz diffusion cell. The results obtained indicated that F5 (3% Thioglycolic acid) and F8 (1.5%Thioglycolic acid + 1.5% DMSO) showed good release. The permeation enhancers showed higher release in the order Thioglycolic acid ? DMSO and combination of permeation enhancers (F16) showed higher release compared to single permeation enhancer.
