THE COMPARATIVE STUDY OF TADALAFIL’S AND DOXAZOSIN’S INFLUENCE ON THE RATE OF OXYGEN CONSUMPTION BY THE TISSUE OF PROSTATE

Abstract

The critical analysis of literature and clinical data gives an opportunity to make the conclusion that intimate molecular mechanisms of clinical effectiveness of doxazosin and tadalafil in men with benign prostatic hyperplasia (BPH) and chronic prostatitis (CP) still remain incompletely understood. We supposed that doxazosin and tadalafil increase the rate of oxygen consumption by the tissue of prostate. The aim is to study how doxazosin and tadalafil impact on the rate of oxygen consumption (ROC) by the homogenates of rats' prostate. Materials and methods: 0.5 liter of distilled water (temperature = 36.6 °С) was poured into two plastic cans. Every can was connected to polarographic device by the pair of electrodes. 66 male rats were used. Under anesthesia the prostate was resected from rats and divided into two parts with equal weight. Homogenate was prepared from every part. Further every homogenate was placed into the cans. The air-tightness was created. The impaction of tadalafil on the ROC was studied in 33 rats. Tadalafil 5 mg was submerged into the first can and none into the second can. The circuit was closed under the voltage of 0.95 V. Current intensity (CI) was the same in both cans in the beginning of every experiment – 1.8 microampere. The synchronous comparison between the CI in two different cans with water in each moment of time was made. The Wilcoxon's criteria was used. It was detected that CI decreases much faster and more remarkably in those cans, where before homogenate immersion 5 mg of tadalafil was placed. Then the impaction of doxazosin 2 mg on the ROC was studied in 33 rats, using the same technology. It was detected that CI decreases much faster and more remarkably in those cans, where before homogenate immersion 2 mg of doxazosin was placed. Conclusion: tadalafil and doxazosin increase the rate of oxygen consumption by the tissue of prostate. In such way, tadalafil and doxazosin help the prostatic tissue to consume О2 supplied by blood. We consider that this is the reason why the clinical efficiency of tadalafil and doxazosin in men with BPH is individual.
